基于调控PI3K-AKT通路探讨艾灸关元穴治疗甲状腺功能减退的作用机制

目的 研究艾灸关元穴对丙硫氧嘧啶(PTU)诱导的甲状腺功能减退(甲减)大鼠的作用,并探讨其潜在的作用机制.方法 将72只Wistar大鼠按照随机数表法分为空白组、模型组和艾灸组,每组24只.模型组和艾灸组大鼠给予PTU灌胃,建立大鼠甲减模型.4周后检测各组大鼠甲状腺功能指标[游离三碘甲状腺原氨酸(FT3)、游离甲状腺素(FT4)、促甲状腺激素(TSH)]水平,造模成功后艾灸组大鼠每隔天在关元穴灸30 min.分别于治疗第4、8、12周腹主动脉取血,检测血清中FT3、FT4及TSH水平.取治疗第12周大鼠血清,观察其对原代大鼠滤泡上皮细胞增殖的影响.RT-PCR检测艾灸对大鼠甲状腺组织中Bcl-2、Bax、Caspase-3、Caspase-9、PI3K p110、AKT1 mRNA表达的影响.结果 与模型组比较,艾灸组大鼠FT3、FT4水平均明显升高(P<0.01),TSH水平明显下降(P<0.01).艾灸组大鼠血清能明显减少大鼠原代甲状腺滤泡细胞的凋亡(P<0.05);艾灸关元穴能明显促进Bcl-2 mRNA的表达(P<0.05),抑制Bax、Caspase-3、Caspase-9 mRNA的表达(P<0.05),促进PI3K p110和AKT1 mRNA表达(P<0.05).结论 艾灸关元穴可明显改善甲减大鼠的甲状腺功能,抑制甲状腺滤泡上皮细胞的凋亡,其机制可能与调节Bcl-2和Bax的平衡以及PI3K-AKT通路有关.
