作者把0.17×10~6个细胞/ml的Madin Darby牛肾细胞悬液植入含有5%胎牛血清单层MEM培养基内,在3%CO_2,37℃条件下培养。植入细胞数为1×10~6个细胞/培皿,细胞存活率用锥虫蓝检查达90%以上。细胞植入24小时后更换新鲜培养基。此时,在培养基中分别加入用磷酸缓冲液(PBS)配制的0.1~10.0μM不同浓度的氯化镉溶液,再培养24小时,检查贴壁细胞数,以此评价氯化镉对牛肾细胞的毒性。
