目的:探讨热休克蛋白70(heat shock protein 70,HSP70)与肿瘤坏死因子-α(tumor necrosis factor-alpha,TNF-α)在甲状腺癌组织中的表达及其与临床病理特征的关系。方法:采用RT-PCR以及Western blot技术检测对比40例甲状腺癌组织、20例甲状腺良性病变组织以及15例正常甲状腺组织中HSP70、TNF-α的表达情况,并分析HSP70表达水平与甲状腺癌临床病理学特征的关系。结果:3组人群组织中HSP70 mRNA水平比较差异有统计学意义(P<0.001),甲状腺癌组织中HSP70 mRNA水平显著高于正常甲状腺组织以及甲状腺良性病变组织(P均<0.05);HSP70 mRNA的表达水平与甲状腺癌组织病理分级显著相关(r s=0.810,P<0.05);甲状腺癌组织中HSP70蛋白水平显著高于正常甲状腺组织(P<0.05);3组人群组织中TNF-αmRNA水平比较差异有统计学意义(P<0.05),甲状腺癌组织中TNF-αmRNA水平显著高于正常甲状腺组织以及甲状腺良性病变组织(P均<0.05)。结论:HSP70、TNF-α在甲状腺癌组织中的表达升高,且HSP70与甲状腺癌组织病理分级明显相关,提示HSP70在甲状腺癌发生发展中发挥重要的作用。
OBJECTIVE: To investigate the expression of heat shock protein 70 (HSP70) and tumor necrosis factor-alpha (TNF-α) in thyroid cancer and its relationship with clinicopathological features. Methods: The expression of HSP70 and TNF-α in 40 cases of thyroid carcinoma, 20 cases of thyroid benign lesions and 15 cases of normal thyroid tissue were detected by RT-PCR and Western blot. The expression of HSP70 was compared with that of thyroid cancer Neo-Confucianism. Results: There was significant difference in the expression of HSP70 mRNA between the three groups (P <0.001). The expression of HSP70 mRNA in thyroid cancer was significantly higher than that in normal thyroid tissue and benign thyroid tissue (P <0.05) (Rs = 0.810, P <0.05). The level of HSP70 protein in thyroid cancer tissues was significantly higher than that in normal thyroid tissues (P <0.05). The levels of TNF-αmRNA in the three groups were significantly different (P <0.05). The level of TNF-αmRNA in thyroid cancer tissues was significantly higher than that in normal thyroid tissues and thyroid benign lesions (all P <0.05). Conclusion: The expression of HSP70 and TNF-α in thyroid cancer tissues is increased, and the expression of HSP70 is significantly correlated with the pathological grade of thyroid cancer, which suggests that HSP70 plays an important role in the occurrence and development of thyroid cancer.
