目的探讨硫化镍诱导人支气管上皮(16HBE)细胞恶性转化作用以及扶正解毒汤(FJD)的保护作用,研究硫化镍致癌的分子机制以及扶正解毒汤对防癌治癌的作用。方法用不同浓度的硫化镍在体外多次处理16HBE细胞,利用刀豆凝集素A(ConA)凝集实验和软琼脂集落形成实验进行转化细胞的恶性鉴定;测定16HBE细胞内镍离子和自由基含量,同时加入扶正解毒含药血清,观察其保护作用。结果硫化镍可诱导16HBE细胞的恶性转化,转化细胞的生长速度加快、排列紊乱、失去接触抑制、呈交叉重叠生长、转化率呈剂量—效应关系。转化细胞可被低浓度ConA凝集并在软琼脂内生长。硫化镍暴露组细胞内镍离子和自由基含量明显增加。中、高剂量的扶正解毒含药血清可通过细胞内、外途径发挥抗氧化、清除自由基效应。结论硫化镍有较强的诱导16HBE细胞恶性转化的能力,具有致癌性。FJD血清对于硫化镍诱导的16HBE细胞恶性转化具有保护作用。
Objective To investigate the malignant transformation of human bronchial epithelial (16HBE) cells induced by nickel sulfide and the protective effect of Fuzheng Jiedu Decoction (FJD). To study the molecular mechanism of nickel sulfide carcinogenesis and the effect of Fuzheng Jiedu Tang on cancer prevention and treatment. Methods Sixteen HBE cells were treated with different concentrations of nickel sulfide in vitro. Cones agglutinin A (ConA) agglutination assay and soft agar colony formation assay were used to identify the malignant cells. The content of nickel ions and free radicals in 16HBE cells was determined. At the same time, adding a detoxification medicine containing serum, observe its protective effect. Results The nickel sulfide could induce the malignant transformation of 16HBE cells. The growth rate of the transformed cells was accelerated, the arrangement was disordered, the contact inhibition was lost, cross overlap growth was observed, and the conversion rate showed a dose-effect relationship. Transformed cells can be agglutinated with low concentrations of ConA and grown in soft agar. Nickel ion and free radical content in the nickel sulfide exposed group increased significantly. Medium and high doses of regurgitation and detoxification drug-containing serum can exert anti-oxidation and scavenging free radical effects through intracellular and extracellular routes. Conclusion Nickel sulfide has a strong ability to induce malignant transformation of 16HBE cells, which is carcinogenic. FJD serum has a protective effect on nickel sulfide-induced malignant transformation of 16HBE cells.
