目的:研究川芎嗪(TMP)对青霉素致痫大鼠神经细胞黏附分子-140(neural cell adhesion molecule-140,NCAM-140)的表达。探讨川芎嗪对青霉素致痫大鼠神经元的保护作用。方法:制备青霉素致痫大鼠模型,将实验大鼠随机分为3组,每组10只:正常对照组,麻醉开颅手术1h后取海马;致痫组,青霉素诱发癫痫1h后取海马;治疗组,青霉素诱发癫痫稳定后,再腹腔注射TMP(40mg.kg-1),待抑制作用最明显时取海马。并用免疫组织化学方法检测神经元内NCAM-140的表达,利用HPIAS-2000图像分析系统测定NCAM-140在以上各组中表达的平均光密度和平均阳性面积率。结果:青霉素诱发癫痫后,腹腔注射TMP(40mg.kg-1)后神经元内NCAM-140的表达较青霉素致痫组高(P<0.05)结论:川芎嗪对青霉素致痫大鼠神经元具有重要的保护作用。
Objective: To investigate the expression of neural cell adhesion molecule-140 (NCAM-140) by ligustrazine (TMP) in penicillin-induced epileptic rats. To investigate the protective effect of ligustrazine on neurons in penicillin-induced epileptic rats. Methods: The rat model of epilepsy induced by penicillin was prepared. The rats were randomly divided into 3 groups (n = 10): normal control group, hippocampus 1h after anesthesia craniotomy, epileptic group, The treatment group, penicillin-induced epilepsy was stable, then intraperitoneal injection of TMP (40mg.kg-1), to be the most obvious inhibition when the hippocampus. The expression of NCAM-140 in neurons was detected by immunohistochemical method. The average optical density and average positive area ratio of NCAM-140 expressed in the above groups were determined by HPIAS-2000 image analysis system. Results: After intraperitoneal injection of TMP (40mg.kg-1), the expression of NCAM-140 in penicillin-induced epilepsy group was higher than that in penicillin-induced epilepsy group (P <0.05) .Conclusion: Tetramethylpyrazine Important protection.
