目的:观察椎动脉型颈椎病(CSA)动物模型血小板聚集功能的改变以及复方川脊片对其的影响。方法:采用硬化剂注射法复制CSA家兔模型,并用复方川脊片治疗,颈复康胶囊作对照,治疗4周后,行经颅多普勒超声(TCD)检查,聚椎动脉行HE染色,测定血管壁中膜厚度,同时采血检测血小板最大聚集率(MAR)。结果:模型组双侧椎动脉和基底动脉血液平均流速(Vm)均较空白组有不同程度降低(P<0.05),动脉的搏动指数(Pi)和阻力指数(Ri)均有不同程度增高(P<0.05);与模型组相比,预复康组和川脊片组上述指标均有不同程度改善(P<0.05),而川脊片组改善更明显。模型组椎动脉管壁中膜平滑肌细胞肥大增生明显,中膜较空白组明显增厚(P<0.05),颈复康组和川脊片组平滑肌细胞增生受到抑制,管壁中膜变薄。模型组血小板MAR较空白组明显增高(R<0.01),颈复康组和川脊片组血小板MAR较模型组明显下降(P<0.05),其中以川脊片组下降更加明显(P<0.01),但是2治疗组血小板MAR仍然明显高于空白组(P<0.05)。结论:复方川脊片可能是通过改善血小板聚集功能,抑制血管壁平滑肌细胞增殖,改善椎-基底动脉血供从而发挥治疗作用。
Objective: To observe the changes of platelet aggregation in animal model of vertebral artery cervical spondylosis (CSA) and the effect of Fufang Chuanji Pian on it. Methods: CSA rabbit model was hardened by injection of sclerotherapy and treated with compound Fufang Chuanji Tablets. After 4 weeks of treatment, transcranial Doppler ultrasound (TCD) The thickness of vessel wall was measured, and the maximum platelet aggregation rate (MAR) was determined by blood sampling. Results: The mean velocity of blood (Vm) in bilateral vertebral artery and basilar artery in model group was lower than that in blank group (P <0.05) and the index of arterial pulsation (Pi) and resistance index (Ri) P <0.05). Compared with the model group, the above indexes in pre-rehabilitation group and chuanji-tablet group improved to some extent (P <0.05), while those in Chuanji-tablet group improved more obviously. The hypertrophic hyperplasia of VSMC in the model group was more obvious than that in the blank group (P <0.05). The proliferation of smooth muscle cells was restrained in the Jinkuikang group and the Chuanji tablet group, and the thickness of the tunica media in the model group was thinner. The platelet MAR of model group was significantly higher than that of blank group (P <0.05), and the decrease of platelet MAR was more significant in model group than that in blank group (P <0.01) ), But the platelet MAR of the 2 treatment groups was still significantly higher than that of the blank group (P <0.05). Conclusion: Fufang Chuanji Tablets may play a therapeutic role by improving platelet aggregation, inhibiting proliferation of vascular smooth muscle cells and improving vertebrobasilar artery blood supply.
