HTLV-1抗体阳性的4例冒烟型ATL(成人T细胞白血病)与5例HTLV-1健康携带者的外周血,用Ficoll-Conray梯度离心,分离单个核细脆并制备涂片(全片通常含有1000个以上靶细胞),经内酮固定(0℃,5分钟)之后,室温下,按下列步骤处理:(1)0.1MPBS(pH7.4,内含0.5%NP-40),30分钟,水洗;(2)鼠单抗FO-120(一种对抗原c-fos原癌基因产物特异的单克隆抗体),40分钟;(3)FIFC标记的抗鼠免疫球蛋白(抗体),40分钟。结果在荧光显微镜下观察,抗原定位于细胞核。
