目的初步建立脑胶质瘤标本库,探讨其应用价值。方法对脑胶质瘤患者进行手术前采静脉血,提取的基因组DNA保存在4℃冰箱内。在肿瘤切除术中,获取部分新鲜肿瘤组织标本和正常脑组织标本,保存在液氮中。收集进行组织病理学检查后剩余的肿瘤组织,制作成石蜡标本保存。在患者术后1年开始对其进行随访。采用生物芯片技术,对与胶质瘤一些相关的分子生物学指标进行初步研究。结果全血中提取的基因组DNA和冻存的肿瘤组织标本中提取的总RNA其 OD260/280比值都在1.7-2.1之间,能够满足胶质瘤分子生物学研究的需要。结论初步建立的脑胶质瘤标本库能够为肿瘤的分子生物学研究提供标本和相关资料来源。
Objective To establish a preliminary brain glioma specimen library and explore its application value. Methods The patients with glioma were collected venous blood before surgery, and the extracted genomic DNA was stored in refrigerator at 4 ℃. In tumor resection, some fresh tumor tissue samples and normal brain tissue samples were obtained and stored in liquid nitrogen. Tumor tissue remaining after histopathological examination is collected and stored as paraffin specimens. Patients were followed up 1 year after surgery. Using biochip technology, some molecular biological indicators related to glioma were preliminary studied. Results The OD260 / 280 ratio of total RNA extracted from whole blood samples and frozen tumor samples was between 1.7 and 2.1, which could meet the needs of molecular biology research of glioma. Conclusion The initial establishment of glioma specimen library can provide samples and relevant data sources for tumor molecular biology research.
