对分离自宁波象山港海水样品中的一株产抗菌肽菌株Y-6进行鉴定。研究了菌株Y-6产抗菌物质对桃软腐菌的抑菌效果。通过形态学特征、生理生化特性以及16S rRNA将菌株Y-6鉴定到种。试验结果表明,Y-6菌株产抗菌肽对桃软腐菌有较强的抑制作用;Y-6菌株为革兰阳性菌,有芽孢居中生长;V-P试验、硝酸盐还原试验、淀粉水解和明胶液化试验、MR、柠檬酸盐利用试验、过氧化氢酶试验都呈阳性,而苯丙氨酸脱氨酶试验呈阴性;该菌能够利用蔗糖、分解酪氨酸,但不能利用麦芽糖和甘露醇;菌株的16S rRNA基因序列和枯草芽孢杆菌接近,其相似性达99.72%。依据《伯杰氏细菌鉴定手册》和16S rRNA同源性比较,将菌株Y-6鉴定为枯草芽孢杆菌。菌株Y-6在桃软腐菌的生物防治中有潜在的应用价值。
An antimicrobial peptide strain Y-6 isolated from seawater samples from Xiangshan Harbor in Ningbo was identified. The antibacterial effect of strain Y-6 producing antibacterial substance on the soft-rot fungi was studied. The strain Y-6 was identified by morphological characteristics, physiological and biochemical characteristics and 16S rRNA. The results showed that the antibacterial peptides produced by strain Y-6 had a strong inhibitory effect on the susceptible fungi Pentaphylococcus aureus. The strain Y-6 was a Gram-positive bacterium, with spores growing centrally. The VP test, nitrate reduction test, starch hydrolysis and gelatin Liquefaction test, MR, citrate use test, catalase test were positive, and phenylalanine deaminase test was negative; the bacteria can use sucrose, tyrosine decomposition, but can not use maltose and mannitol The 16S rRNA gene sequence of Bacillus subtilis was similar to that of Bacillus subtilis, and its similarity was 99.72%. The strain Y-6 was identified as Bacillus subtilis according to the Berger’s Handbook of Bacteriology Identification and 16S rRNA homology comparison. The strain Y-6 has potential application value in biological control of peach soft rot.
