采用银染PCR-SSCP方法检测NNK诱发BEP2D恶性转化细胞中p16基因变化。与对照组比较,转化细胞中p16基因的E1.β外显子带型出现变异,而第1~3外显子均未见泳动变位,提示E1.β外显子突变和/或缺失所致细胞周期失调是NNK诱发人类支气管上皮细胞癌变的可能原因之一。
The silver staining PCR-SSCP method was used to detect the changes of p16 gene in malignant transformed BEP2D cells induced by NNK. Compared with the control group, E1 of the p16 gene in the transformed cells. There was a variation in the beta exon type, and no migration was seen in exon 1 to exon 3, suggesting E1. Dysregulation of cell cycle due to mutations and/or deletions of exon β is one of the possible causes of NNM-induced carcinogenesis of human bronchial epithelial cells.
