目的:探索D101大孔吸附树脂制备半边旗中抗肿瘤有效成分贝壳杉烷类二萜5F的工艺。方法:以HPLC为检测手段,以5F在浸膏中的含量为指标,比较甲醇和氯仿提取5F的专属性;以HPLC和TLC为检测手段,考察D101大孔吸附树脂对5F的吸附和洗脱条件。结果:半边旗药粉采用氯仿提取,制成含乙醇20%的药液上柱后,先用2BV30%的乙醇除去极性杂质,再用4BV60%的乙醇洗脱,得到质量分数为32.25%的5F。结论:建立了一种5F的纯化工艺,并达到要求的标准。
Objective: To explore the preparation of anti-tumor effective component kaurizan diterpenoid 5F from D101 macroporous resin. Methods: HPLC was used as a detection method and the content of 5F in the extract was used as an index to compare the specificity of 5F with methanol and chloroform. The adsorption and elution of 5F by D101 macroporous resin was investigated by HPLC and TLC. condition. RESULTS: The Fructus regale herbicide powder was extracted with chloroform and made into a solution containing ethanol 20%. After removing the polar impurities with 2BV 30% ethanol and then eluting with 4BV 60% ethanol, a 5F mass fraction of 32.25% was obtained. . Conclusion: A 5F purification process was established and met the required standards.
