目的以糖尿病大鼠为实验对象,研究单纯高血糖是否可诱发动脉粥样硬化,以及游离脂肪酸在糖尿病大血管动脉粥样硬化中的作用。方法用形态学方法观察糖尿病大鼠动脉粥样硬化的表现;抽血化验游离脂肪酸、血脂和血糖水平;用免疫组织化学方法研究动脉粥样硬化局部的炎症介质表达。结果实验第3个月时光镜下可见动脉血管壁局限增厚,可观察到泡沫细胞;第5个月时已形成脂纹。糖尿病各组与对照组比较,血糖、甘油三酯和游离脂肪酸水平升高(P<0.05),一氧化氮水平降低(P<0.05)。免疫组织化学显示,白细胞介素6和肿瘤坏死因子α在糖尿病大鼠血管内膜、中膜层细胞的胞浆及细胞间质均有表达。结论单纯高血糖动物模型出现了明确的动脉粥样硬化部分特征性表现,说明高血糖是动脉粥样硬化的另一重要原因;游离脂肪酸增高可能是糖尿病动脉粥样硬化的危险因素,其作用可能与动脉壁局部炎症介质表达有关。
Objective To investigate whether hyperglycemia can induce atherosclerosis and the role of free fatty acids in atherosclerosis of diabetic macroangiopathy in diabetic rats. Methods Morphological methods were used to observe the atherosclerosis in diabetic rats. Free fatty acids, lipids and blood glucose levels were determined by blood chemistry. The expression of inflammatory mediators in atherosclerotic lesions was studied by immunohistochemistry. Results In the third month of the experiment, arterial wall thickening was observed under the light microscope, and foam cells were observed. At the fifth month, lipid streaks were formed. Compared with the control group, the level of blood glucose, triglyceride and free fatty acid increased (P <0.05) and the level of nitric oxide decreased (P <0.05). Immunohistochemistry showed that interleukin-6 and tumor necrosis factor-alpha were expressed in the cytoplasm and interstitial cells of vascular intima and media of diabetic rats. Conclusion A clear hyperlipidemia animal model showed a clear atherosclerotic part of the characteristic manifestation of hyperglycemia is another important cause of atherosclerosis; increased free fatty acids may be atherosclerosis in diabetic risk factors, its role may be And arterial wall local inflammatory mediators related.
