采用裸鼠移植人肝癌模型,观察rhTNF对肝癌细胞的抑制作用。结果发现rhTNF瘤内注射或腹腔注射对肝癌细胞均有抑制作用;高剂量组(10000U/只)和中剂量组(2000U/只)对肿瘤体积的抑制率分别为33.4%和25.1%(P<0.05~0.01),10000U/只剂量的rhTNF腹腔注射其抑制率为31.6%(P<0.05);瘤体重量的抑制在高剂量组、中剂量组和腹腔组分别为39%、33%、29.7%。病理检查发现高剂量组和中剂量组瘤组织中央部位有程度不等的出血坏死区域。
Human hepatocellular carcinoma model was implanted in nude mice to observe the inhibitory effect of rhTNF on hepatoma cells. The results showed that intratumoral injection of rhTNF or intraperitoneal injection had inhibitory effects on hepatoma cells; the inhibitory rates of tumor volume were 33.4% and 25.1 in the high-dose group (10000 U/dose) and the middle-dose group (2000 U/dose), respectively. % (P<0.05-0.01). The inhibitory rate of intraperitoneal injection of 10000U/dose of rhTNF was 31.6% (P<0.05); inhibition of tumor weight was seen in high-dose group and middle-dose group. In the celiac group, they were 39%, 33%, and 29.7%, respectively. Pathological examination revealed that there were varying degrees of hemorrhage and necrosis in the central parts of the tumor tissue in the high dose group and the middle dose group.
