目的 :观察血管内皮生长因子受体 flt- 1和 KDR在骨肿瘤中的表达 ,探讨 flt- 1和 KDR与骨肿瘤的生长、恶性程度及临床病理关系。方法 :采用免疫组化 SP法染色和图像分析系统检测 90例骨肿瘤中 flt- 1和 KDR的表达。结果 :2 0例骨软骨瘤 flt- 1和 KDR染色均为阴性。在骨巨细胞瘤及骨肉瘤中 flt- 1和 KDR均有表达。 flt- 1和 KDR不仅在血管内皮细胞中 ,在部分肿瘤细胞也呈阳性反应。骨肉瘤中 flt- 1和 KDR的表达明显高于骨巨细胞瘤 (P <0 .0 5 )。在骨巨细胞瘤中 ,随着分化程度的下降 ,受体的表达有逐渐递增的趋势。 、 级与 级之间存在显著性差异 (P <0 .0 5 )。 级与 级之间无差异 (P >0 .0 5 )。相关分析显示 flt- 1和 KDR的表达与骨巨细胞瘤的病理分级密切相关。结论 :VEGF及其受体 flt- 1和 KDR影响肿瘤细胞的生长及间质中新生血管形成 ,肿瘤细胞的生长和血管的发生相互依赖。 flt- 1和 KDR的表达与骨肿瘤的恶性程度密切相关。恶性程度越高 ,表达越增强。 flt- 1和 KDR在骨肿瘤中的表达可能作为判断骨肿瘤恶性度的一项有价值的指标。
Objective : To observe the expression of vascular endothelial growth factor receptor flt-1 and KDR in bone tumors, and to investigate the growth, malignancy and clinicopathological relationship between flt-1 and KDR and bone tumors. Methods : The expression of flt-1 and KDR in 90 cases of bone tumors was detected by immunohistochemical SP staining and image analysis system. Results : Twenty cases of osteochondroma had negative flt-1 and KDR staining. Both flt-1 and KDR are expressed in giant cell tumor of bone and osteosarcoma. Flt-1 and KDR are not only positive in vascular endothelial cells but also in some tumor cells. The expression of flt-1 and KDR in osteosarcoma was significantly higher than that of giant cell tumor of bone (P < 0.05). In giant cell tumor of bone, as the degree of differentiation decreases, the expression of receptors gradually increases. There was a significant difference between the grade, grade and grade (P < 0.05). There is no difference between grades and grades (P > 0.05). Correlation analysis showed that the expression of flt-1 and KDR was closely related to the pathological grade of giant cell tumor of bone. Conclusion: VEGF and its receptors flt-1 and KDR affect the growth of tumor cells and neovascularization in the interstitium. The growth of tumor cells and the occurrence of blood vessels depend on each other. The expression of flt-1 and KDR is closely related to the degree of malignancy of bone tumors. The higher the degree of malignancy, the stronger the expression. The expression of flt-1 and KDR in bone tumors may be a valuable indicator to determine the degree of malignancy of bone tumors.
