目的:观察清毒伸筋汤对胶原诱导性关节炎大鼠血清肿瘤坏死因子α(tumor necrosis factorα,TNF-α)、白细胞介素-1β(interleukin-1β,IL-1β)的影响,从分子水平探讨清毒伸筋汤治疗类风湿关节炎的作用机理。方法:60只大鼠随机选取10只为正常组,其余大鼠采用弗氏不完全佐剂和牛II型胶原蛋白诱导大鼠类风湿关节炎模型,模型制作成功后随机分为模型组,甲氨蝶呤组,清毒伸筋汤高剂量组、中剂量组、小剂量组。各治疗组灌服相应药物,正常组和模型组给予生理盐水。28 d后,各组小鼠主动脉采血,采用酶联免疫吸附法检测各组大鼠TNF-α、IL-1β水平。结果:清毒伸筋汤各剂量组均能降低大鼠血清TNF-α、IL-1β的水平,其中高剂量组、中剂量组与模型组比较,差异有统计学意义(P<0.05或P<0.01)。清毒伸筋汤中剂量与甲氨蝶呤组比较,差异无统计学意义(P>0.05),两者效果相当。结论:清毒伸筋汤能降低类风湿关节炎大鼠血清中TNF-α、IL-1β水平,从而抑制炎症反应,进一步阻止骨质破坏。
Objective: To observe the effect of Qingdu shenjin decoction on serum tumor necrosis factorα (TNF-α) and interleukin-1β (IL-1β) in collagen-induced arthritis rats, To explore the mechanism of Qingdu shenjin decoction in treating rheumatoid arthritis. Methods: Sixty rats were randomly selected as the normal control group. The rest rats were induced with Freund’s incomplete adjuvant and bovine type II collagen to induce rheumatoid arthritis in rats. The model rats were randomly divided into model group, Pterin group, Qingdu shenjin decoction high dose group, middle dose group, low dose group. The treatment groups were fed the corresponding drugs, normal group and model group given saline. After 28 days, blood was collected from the aorta in each group, and the levels of TNF-α and IL-1β in each group were detected by enzyme-linked immunosorbent assay. Results: The Qingdu shenjin decoction could reduce the levels of serum TNF-α and IL-1β in rats, and there was significant difference between high dose group and middle dose group and the model group (P <0.05 or P <0.01). There was no significant difference between the moderate dose of Qingdu Soup in Methotrexate group and Methotrexate group (P> 0.05). Conclusion: Qingdu Shenjin Decoction can reduce the levels of TNF-α and IL-1β in the serum of rheumatoid arthritis rats, thereby inhibiting the inflammatory reaction and further preventing the bone destruction.
