目的研究骨髓间充质干细胞(MSCs)移植治疗对Duchenne肌营养不良(DMD)模型鼠mdx鼠病态肌肉的影响。方法分离培养正常小鼠MSCs,局部肌肉注射移植入DMD型模型鼠mdx鼠肌肉,16周后观察mdx鼠肌肉肌力、肌肉病理学改变及dystrophin蛋白表达变化。结果MSCs移植16周后mdx鼠肌纤维dystrophin蛋白表达增加,肌力好转,核中移明显改善。结论MSCs移植可缓解mdx鼠肌肉病理改变,但移植效率的提高需要进一步研究。
Objective To study the effect of bone marrow mesenchymal stem cells (MSCs) transplantation on the morbid muscle of mdx mice with Duchenne muscular dystrophy (DMD). Methods MSCs of normal mice were isolated and cultured. Mdx mice were transplanted intramuscularly into DMD model muscle. Muscle strength, muscle pathology and expression of dystrophin protein in mdx mice were observed after 16 weeks. Results MSCs transplantation 16 weeks after mdx mouse muscle fiber dystrophin protein expression increased, muscle strength improved, the nuclear shift significantly improved. Conclusion MSCs transplantation can alleviate the muscle pathological changes in mdx mice, but the improvement of transplantation efficiency needs further study.
