人的脐带血清中抗体介导ADCC的作用已为大家所熟知,本文作者最近用11例正常新生儿分娩过程中收集的脐带血和7例健康成年供血者的外周血,经聚蔗糖—泛影葡胺梯度分离出单个核细胞。将单个核细胞置于含有小牛血清的塑料板内,37℃2小时分离出粘附与非粘附淋巴细胞。用~(51)Cr释放研究ADCC对流感病毒A/ALask/6/77(H+3N+2)感染细胞溶解作用。通过间接免疫荧光鉴定法计数与HNK—1抗体反应的细胞。作者认为,脐带血淋巴细胞的ADCC活性比成年人淋巴细胞的ADCC活性明显的低。
The role of antibody-mediated ADCC in human umbilical cord serum has been well known. The authors recently used peripheral blood from cord blood collected from 11 normal newborn infants during delivery and from 7 healthy adult donors, Mono-myeloid gradient isolation of mononuclear cells. Mononuclear cells were placed in plastic plates containing bovine serum and adherent and non-adherent lymphocytes were isolated at 37 ° C for 2 hours.
