目的探讨碳酸锂 (Li2 CO3 )对抗黄曲霉素B1(AFB1)诱导大鼠肝癌前病变过程中P2 1ras、P2 1WAF1蛋白的表达及意义。方法免疫组织化学方法 (ABC法 )。结果P2 1ras蛋白在实验第 6周各组表达增加 ,P2 1WAF1蛋白则表达减少 ,均早于病理组织学改变 ;第 10周时P2 1ras显著增高 ,而P2 1WAF1改变不明显 ,两指标呈负相关关系 ;LiCO3 同时给药组、Li2 CO3 先期给药组组织形态学改变明显好于阳性对照组 ,肝癌前病变程度轻 ,变化趋势为阳性对照组→LiCO3 同时给药组→Li2 CO3 先期给药组→阴性对照组。结论Li2 CO3 可对抗AFB1诱导的大鼠肝癌的发生发展 ,可明显抑制P2 1ras的过度表达和促进P2 1WAF1的调控作用 ;P2 1ras、P2 1WAF1蛋白的动态检测有助于肝癌的早期发现及预后的判断。
Objective To investigate the expression and significance of P2 1ras and P2 1WAF1 protein in the precancerous lesions of rat hepatoma induced by aflatoxin B1 (AFB1) with lithium carbonate (Li2CO3). Methods Immunohistochemical methods (ABC method). Results The expression of P2 1ras protein was increased in each group at the 6th week, and the expression of P2 1WAF1 protein was decreased, which was earlier than histopathological changes. At the 10th week, P2 1ras increased significantly, but P2 1WAF1 did not change significantly. The two indicators were negatively correlated. Relationship; LiCO3 co-administration group, Li2CO3 pre-administration group, histological changes were significantly better than the positive control group, the precancerous lesions of liver were mild, the trend of change was positive control group → LiCO3 simultaneous administration group → Li2CO3 pre-administration group → Negative control group. Conclusion Li2CO3 can inhibit the occurrence and development of rat hepatoma induced by AFB1, and can significantly inhibit the overexpression of P2 1ras and promote the regulation of P2 1WAF1. The dynamic detection of P2 1ras and P2 1WAF1 protein contributes to the early detection and prognosis of liver cancer. judgment.
