目的了解漳州市肠道病毒71型分离株的VP1区基因特征。方法对2010年漳州市的5株肠道病毒71型分离株进行VP1区基因全长序列测定,测序结果利用DNASTAR软件进行核苷酸、氨基酸序列分析和同源性比较,并用Mega4.0软件构建系统发生树。结果 5株肠道病毒71型分离株的VP1区核苷酸序列全长均为891bp,且与2008年安徽阜阳流行株Fuyang.Anhui.P.R.C-17.08-2的VP1区核苷酸序列同源性和氨基酸序列同源性最高,分别为98.2%~98.8%和99.7%~100.0%。5株肠道病毒71型分离株间的核苷酸序列同源性为97.2%~99.8%,氨基酸序列同源性为99.3%~100.0%。氨基酸在98位和218位这两个位点发生了特异性变异。VP1区基因遗传进化分析表明,漳州市所有分离株均属于C4基因亚型的C4a进化分支。结论 5株肠道病毒71型分离株均属于C4基因亚型的C4a进化分支,与2004年以来的中国大陆EV71病毒流行的基因型完全一致。
Objective To understand the characteristics of VP1 gene of enterovirus 71 strain in Zhangzhou City. Methods The full length sequence of VP1 gene of five enterovirus 71 isolates in Zhangzhou City in 2010 was determined. The nucleotide and amino acid sequences were analyzed by DNASTAR software and the homology was compared. The results of Mega4.0 software System occurrence tree. Results The total length of the VP1 region of the 5 enterovirus 71 isolates was 891 bp, which was identical to the nucleotide sequence of the VP1 region of Fuyang.Anhui.PRC-17.08-2 in Fuyang, Anhui province in 2008 It has the highest homology with the amino acid sequence of 98.2% -98.8% and 99.7% -100.0%, respectively. The nucleotide sequence homologies among the five enterovirus 71 isolates ranged from 97.2% to 99.8% with homology of 99.3% to 100.0%. Amino acids at 98 and 218 of these two sites of a specific mutation occurred. Genetic analysis of VP1 gene showed that all the isolates in Zhangzhou belonged to the C4a evolutionary branch of C4 subtype. Conclusion All five enterovirus 71 isolates belonged to the C4a evolutionary branch of C4 subtype, which was completely consistent with the genotype of EV71 virus in mainland China since 2004.
