目的探讨抗精子抗体(AsAb)阳性大鼠精子凋亡率的变化及其可能的机制。方法建立AsAb阳性大鼠动物模型,HE染色观察大鼠附睾组织形态;采用3’-原位末端DNA标记(TUNEL)技术检测大鼠精子的凋亡率;应用化学比色法测定精子匀浆的MDA含量和总抗氧化能力;应用定磷法测定精子匀浆的Na+-K+-ATP酶和Ca2+-ATP酶活性。结果与正常组相比,AsAb阳性组大鼠精子凋亡率和MDA含量显著增加(P<0.01;P<0.01);总抗氧化能力、Na+-K+-ATP酶和Ca2+-ATP酶活性显著降低(P<0.01;P<0.01;P<0.05)。结论AsAb阳性大鼠精子凋亡率增加;可能与氧自由基含量增多、Na+-K+-ATP酶和Ca2+-ATP酶活性降低有关。
Objective To investigate the changes of sperm apoptosis rate in AsAb positive rats and its possible mechanism. Methods The animal model of AsAb positive rat was established and the morphology of epididymis was observed by HE staining. The apoptosis rate of spermatozoa was detected by 3’-TUNEL. The chemical colorimetry was used to determine the sperm homogenate MDA content and total antioxidant capacity. The activities of Na + -K + -ATPase and Ca2 + -ATPase in sperm homogenate were determined by fixed phosphorus method. Results Compared with the normal control group, the apoptosis rate and the content of MDA in AsAb positive group were significantly increased (P <0.01; P <0.01). The total antioxidant capacity, Na + -K + -ATPase and Ca2 + -ATPase activity were significantly decreased (P <0.01; P <0.01; P <0.05). Conclusion AsAb positive rat sperm apoptosis increased; may be related to increased oxygen free radicals, Na + -K + -ATPase and Ca2 + -ATPase activity.
