目的：通过比较三种从石蜡组织中提取DNA的不同方法，寻找一种高效、经济而简便的方法指导实际工作。方法选取结外淋巴组织增生性病变8例，采取三种方法提取DNA,分别是酚-氯仿提纯法，快速法和试剂盒法。通过检测提取DNA的浓度和纯度，PCR扩增效果，比较三种DNA提取方法的优劣。结果试剂盒法提取DNA OD260/280比值在1.91-1.96间；酚-氯仿提纯法OD值在1.32-1.68间；快速法OD值在1.19-1.36间。其中试剂盒提取DNA最为稳定。3种方法提取的DNA为模板行PCR扩增，阳性率无统计学意义。结论快速法简单、无毒、需要的样品少，是一种值得推荐的DNA提取方法。“,”[abstract] objective to seek an effective economic and simple DNA Extraction methods from Unbufered Fomalirr-Fixed and paraffin-Embedded tissue ,three chosen methods were employed in our study. Methods 8 blocks of lymphoid tissue were used in the test. Three chosen methods respectively are simply digestion and boiling,phenol/chloroform purification, and commercial Kit methods. Results Kit law to extract DNA OD260/280 ratio between 1.91 1.96; Phenolic-chloroform purification method to get DNAOD260/280 ratio between 1.32 1.68; Quick method to get DNAOD260/280 ratio between 1.19 1.36. One kit extract DNA most stable. Three kinds of methods to extract DNA for template amplification, PCR positive was not statisticaly significant.Conclusion Rapid method is a recommended DNA extraction method. Because it is simple, non-toxic and need less samples.
