目的 观察糖皮质激素对骨髓基质细胞增殖及分化的作用,探讨糖皮质激素导致继发性骨质疏松的病理学机制。方法 以1×10-7M地塞米松加入骨髓基质细胞培养物中,测定增殖的骨髓基质细胞数及培养液中骨钙素含量。通过苏丹Ⅲ脂肪细胞染色计数观察地塞米松作用时间对脂肪细胞分化的影响。结果 实验组骨髓基质细胞数及培养液中骨钙素含量明显低于对照组,脂肪细胞的数量随地塞米松作用时间延长而增多。结论 糖皮质激素抑制骨髓基质细胞增殖及向成骨细胞分化,促进其向脂肪细胞分化,这可能与糖皮质激素引起继发性骨质疏松时骨量减少、髓内脂肪组织增多有关。
Objective To observe the effect of glucocorticoids on the proliferation and differentiation of bone marrow stromal cells and to explore the pathological mechanism of glucocorticoid-induced secondary osteoporosis. Methods 1 × 10-7M dexamethasone was added into bone marrow stromal cell culture to determine the number of proliferating bone marrow stromal cells and osteocalcin in culture medium. The effect of dexamethasone on adipocyte differentiation was observed by Sudan III staining of adipocytes. Results The number of bone marrow stromal cells and osteocalcin in experimental group were significantly lower than those in control group. The number of adipocytes increased with the prolongation of dexamethasone treatment. Conclusion Glucocorticoid inhibits the proliferation of bone marrow stromal cells and differentiates into osteoblasts, and promotes their differentiation into adipocytes. This may be related to the decrease of bone mass and the increase of intra-adipose tissue when glucocorticoid-induced secondary osteoporosis.
