目的动态观测脑出血过程中大鼠血肿周边、下丘脑及脑干NPY含量的变化,探讨脑出血过程中中枢神经系统神经肽Y(NPY)含量的动态变化及意义。方法采用胶原酶和肝素联合注入尾状核的方法建立大鼠脑出血模型,测定出血前、出血后30min,6,12,24,48及72h血肿周边、下丘脑及脑干NPY的活性。NPY活性的测定采用放射免疫法。结果脑出血后血肿周边、下丘脑及脑干NPY的含量同步升高,并于24h达峰值,分别为犤(4.37±0.14),(9.06±0.15),(7.22±0.26)μg/gprotein犦,48~72h开始回落。但仍显著高于出血前(t=31.3216,12.5715,11.6487,P<0.01)。结论脑出血过程中,脑组织NPY活性增强,导致局部脑血流量减少,最终加重神经细胞损害。
Objective To observe the changes of NPY contents in the perihematoma, hypothalamus and brainstem during cerebral hemorrhage and to explore the dynamic changes and significance of neuropeptide Y (NPY) content in central nervous system during intracerebral hemorrhage. Methods A rat model of intracerebral hemorrhage was established by injecting collagenase and heparin into the caudate nucleus. The activity of NPY in perihematoma, hypothalamus and brainstem was measured at 30, 6, 12, 24, 48 and 72 h after hemorrhage. NPY activity was measured by radioimmunoassay. Results The content of NPY in perihematoma, hypothalamus and brainstem after ICH increased synchronously and reached the peak value at 24h (37 4.37 ± 0.14, (9.06 ± 0.15), (7.22 ± 0.26) μg / gprotein 犦, 48 ~ 72h began to fall. But still significantly higher than before bleeding (t = 31.3216,12.5715,11.6487, P <0.01). Conclusions During the process of intracerebral hemorrhage, the activity of NPY in the brain tissue is enhanced, which leads to the decrease of the local cerebral blood flow and eventually aggravates the damage of nerve cells.
