目的了解诱导性多能干细胞(induced pluripotent stem cells,IPS细胞)对肝细胞损伤的治疗作用。方法取健康雄性C57小鼠28只,分为健康对照组、实验组和实验对照组。实验组小鼠腹腔内注射2500 mg/kg氨基半乳糖造小鼠肝功能损伤模型,造模2天后在实验组小鼠尾静脉注射CFSE荧光标记后的IPS细胞。在注射IPS细胞后5天、10天、15天和20天时,健康对照组、实验组、实验对照组各取两只小鼠处死,取肝脏标本,分别行冰冻切片显微镜观察荧光,石蜡切片行HE染色,取血行生物化学检查。结果IPS细胞注射入小鼠体内后,可在小鼠肝脏定植,注射IPS细胞的小鼠血中ALT、AST下降速度较未注射IPS细胞的小鼠明显加快。肝脏病理损伤恢复情况较未注射IPS细胞的小鼠明显改善。结论 IPS细胞注射入肝功能损伤C57小鼠体内,可对小鼠的肝功能损伤有一定的修复作用。
Objective To investigate the therapeutic effect of induced pluripotent stem cells on hepatocellular injury. Methods Twenty-eight healthy male C57 mice were divided into healthy control group, experimental group and experimental control group. The experimental mice were injected intraperitoneally with 2500 mg / kg galactosamine to establish mice model of hepatic injury. Two days later, CFSE fluorescent labeled IPS cells were injected into tail vein of experimental mice. At 5 days, 10 days, 15 days and 20 days after the IPS cells were injected, two mice in the healthy control group, the experimental group and the experimental control group were sacrificed and the liver samples were taken. The frozen sections were observed under fluorescence microscope and paraffin sections HE staining, blood biochemical examination. Results IPS cells were injected into mice and colonized in the liver of mice. The reduction of ALT and AST in mice injected with IPS cells was significantly faster than that in mice without IPS cells. The recovery of hepatic pathological injury was significantly improved than that of mice without IPS cells. Conclusion The injection of IPS cells into C57 mice with hepatic impairment can repair the liver injury in mice.
