目的 研究阻断泛素 -蛋白酶体通路对胃癌细胞株SGC -790 1DNA合成及细胞周期的影响。方法 将泛素 -蛋白酶体通路特异性阻断剂MG -13 2加入胃癌细胞株SGC -790 1,3 H -TdR掺入试验测定对胃癌细胞DNA合成的影响 ,流式细胞仪(FCM)检测细胞周期。结果 MG -13 2作用于胃癌细胞株SGC -790 1后 ,细胞DNA合成显著受抑制 ,G0 /G1 期细胞比例增加 ,S、G2 /M期细胞比例减少。结论 MG -13 2能显著抑制胃癌细胞株SGC -790 1的DNA合成 ,使细胞产生G1 期阻滞 ,表明阻断泛素 -蛋白酶体通路可能是治疗胃癌的新思路
Objective To investigate the effects of blocking ubiquitin-proteasome pathway on DNA synthesis and cell cycle of gastric cancer cell line SGC-790.1. Methods The effects of ubiquitin-proteasome pathway-specific inhibitor MG-132 on gastric cancer cell line SGC-7901 and 1,3 H -TdR were tested by flow cytometry (FCM) cell cycle. Results After treated with MG-132, the DNA synthesis of gastric cancer cell line SGC-7901 was significantly inhibited. The proportion of cells in G0 / G1 phase increased and the proportion of cells in S, G2 / M phase decreased. Conclusion MG-132 can significantly inhibit the DNA synthesis of gastric cancer cell line SGC-7901 and cause cell arrest in G1 phase, indicating that blocking the ubiquitin-proteasome pathway may be a new way of treating gastric cancer
