目的:观察牛蒡子苷元对S180荷瘤小鼠肿瘤组织生长及其血管生成的影响。方法:以S180荷瘤小鼠为研究对象,设置阴性对照组(溶剂)、阳性药物组5-氟尿嘧啶40mg/kg和牛蒡子苷元低、中、高剂量组(40、80、160 mg/kg),按0.01 ml/g,每日灌胃给药1次,连续10 d,脱颈椎处死小鼠,对肿瘤组织及脾脏称重并计算抑瘤率及脾脏指数,用免疫荧光染色法检测肿瘤组织内微血管密度(MVD)和血管生长因子VEGF的表达。结果:牛蒡子苷元不同剂量组对S180荷瘤小鼠肿瘤的生长和肿瘤组织中的微血管密度(MVD)及VEGF的表达均有显著的抑制作用,牛蒡子苷元40、80、160 mg/kg的抑瘤率分别为23.00±13.10%,40.42±12.90%和45.67±16.15%,肿瘤组织微血管密度(MVD)分别为38.25±4.53、32.62±3.66和22.75±5.28,免疫组化染色VEGF相对光密度值分别为75.66±8.41%、70.32±5.73%和60.66±9.38%。结论:牛蒡子苷元能够抑制S180荷瘤小鼠肿瘤生长及其瘤内微血管密度,对肿瘤组织内VEGF的表达亦有抑制作用。
Objective: To observe the effect of arctigenin on tumor growth and angiogenesis in S180 tumor-bearing mice. Methods: S180 tumor-bearing mice were used as experimental subjects. Negative control group (solvent), positive control group, 5-fluorouracil 40 mg / kg and arctigenin low, medium and high dose groups (40, 80, 160 mg / kg ), By 0.01 ml / g, administered orally once a day for 10 days.
