目的:探讨虎金颗粒对肝纤维化相关炎症细胞因子中白细胞介素-1(IL-1β)、肿瘤坏死因子-α(TNF-α)和转化生长因子β1(TGF-β1)的影响。方法:猪血清腹腔注射诱导免疫性肝纤维化大鼠模型,不同剂量(2.4、1.2、0.6 g/kg)的虎金颗粒干预后,采用放射免疫法(RIA)测定大鼠血清中IL-1β、TNF-α的含量,并通过逆转录聚合酶链反应(RT-PCR)半定量法检测大鼠肝组织中TGF-β1mRNA的表达。结果:与模型组比较,虎金颗粒组大鼠血清中IL-1β、TNF-α的含量及肝组织中TGF-β1mRNA表达显著降低(P<0.01)。结论:虎金颗粒可通过抑制炎症细胞因子IL-1β、TNF-α和TGF-β1的表达而起到抗肝纤维化作用。
Objective: To investigate the effect of Hujin Granule on interleukin-1 (IL-1β), tumor necrosis factor-α (TNF-α) and transforming growth factor β1 (TGF-β1) in inflammatory cytokines related to liver fibrosis. Methods: The rat model of autoimmune liver fibrosis was induced by intraperitoneal injection of porcine serum. After intervention of Hujin Granules (2.4, 1.2, 0.6 g / kg) at different doses, the levels of IL-1β , And TNF-α. The expression of TGF-β1 mRNA in rat liver tissue was detected by reverse transcription-polymerase chain reaction (RT-PCR) semiquantitative method. Results: Compared with model group, the contents of IL-1β and TNF-α in serum and the expression of TGF-β1 mRNA in liver tissue of Hujin Granule group were significantly decreased (P <0.01). Conclusion: Hujin Granule can prevent hepatic fibrosis by inhibiting the expression of inflammatory cytokines IL-1β, TNF-α and TGF-β1.
