目的:了解肝枯否细胞膜结合型肿瘤坏死因子(TNF)在严重烫伤后肝损伤中的作用。方法:建立大鼠30%体表面积的全层皮肤烫伤模型,应用流式细胞术对烫伤后枯否细胞膜结合型TNF的表达与肝功能损害的关系进行研究。结果:烫伤可明显增加受内毒素刺激的枯否细胞分泌型TNF的量。烫伤后枯否细胞膜结合型TNF的表达明显升高,但其对内毒素的反应性已不明显。烫伤后枯否细胞膜结合型TNF的表达与血浆谷丙转氨酶改变呈正相关(r=0.902,P<0.001)。结论:烫伤后肝枯否细胞膜结合型TNF与分泌型TNF的意义不尽相同,肝枯否细胞膜结合型TNF在烫伤后肝损伤的发生中起着重要的作用
Objective: To investigate the role of hepcidin cell membrane bound tumor necrosis factor (TNF) in the pathogenesis of severe postburn liver injury. Methods: A full-thickness skin scald model with 30% body surface area was established in rats. The relationship between the expression of membrane-bound TNF in Kupffer cells and liver dysfunction was studied by flow cytometry. Results: Scald significantly increased endotoxin-stimulated Kupffer cell secretion of TNF. After scald Kupffer cell membrane-bound TNF expression was significantly increased, but its reactivity to endotoxin is not obvious. There was a positive correlation between the expression of plasma membrane bound type TNF and plasma alanine aminotransferase (r = 0.902, P <0.001) after scald. CONCLUSION: The significance of cell membrane bound TNF and secreted TNF in Hepatic Fibroblasts after burn is different. Hepatic Fibroblasts cell membrane bound TNF plays an important role in the occurrence of postburn liver injury
