本文以草莓森嘎拉组培苗离体叶片为外植体,研究了暗培养、不同激素配比对不定芽分化及暗培养对叶片不定芽诱导率的影响。结果表明:森嘎拉的叶片诱导最适培养基为MS+3.0 mg/L 6-BA+0.2 mg/L2,4-D,在此培养基上诱导率可达73.3%,平均出芽数可达2.2。先暗培养7 d,再进行光培养,不定芽的诱导率较高,不定芽诱导率为78.2%。对卡那霉素筛选压力和除菌剂浓度的研究表明:用300 mg/L(Cef)除菌,外植体分化率最高,并确立了森嘎拉在不定芽分化阶段的Km筛选压力为20 mg/L。
In this paper, the isolated leaves of strawberry seedlings were used as explants to study the effects of dark and different hormones on adventitious bud induction of adventitious buds. The results showed that the optimum induction medium for the induction of the leaves of the seedlings was MS + 3.0 mg / L 6-BA + 0.2 mg / L 2,4-D. The induction rate on the medium was 73.3% 2.2. The first dark culture 7 d, and then light culture, adventitious buds higher induction rate, adventitious bud induction rate was 78.2%. The results of kanamycin screening pressure and concentration of antibacterial agent showed that the rate of explant differentiation was the highest with 300 mg / L (Cef) sterilization, and the Km selection pressure for the stage of adventitious bud differentiation 20 mg / L.
