目的:观察5-羟色胺对大鼠新鲜分离背根神经节激活电流的调制作用。方法:取大鼠背根神经节,经酶消化,分离神经元,应用全细胞膜片钳技术,记录细胞膜电流。结果:在部分细胞(24/40)5-羟色胺(10-4~10-7mol/L)可引起浓度依赖性的内向电流;部分细胞(12/40)未检测到膜电流的变化;部分细胞(4/40)可引起微弱的外向电流。预加5-羟色胺(10-4~10-8mol/L),对γ-氨基丁酸激活电流明显抑制且呈剂量依赖性。结论:5-羟色胺可抑制γ-氨基丁酸激活电流。
OBJECTIVE: To observe the modulating effect of 5-hydroxytryptamine on freshly isolated DRG activation current in rats. Methods: The dorsal root ganglion (DRG) of rats was taken and neurons were isolated by enzyme digestion. The whole cell patch clamp technique was used to record the cell membrane current. Results: In some cells (24/40), serotonin (10-4 ~ 10-7mol / L) induced a concentration-dependent inward current; no change of membrane current was observed in some cells (12/40) (4/40) can cause weak outward current. Serotonin pretreatment (10-4 ~ 10-8mol / L), the γ-aminobutyric acid activation current was significantly inhibited and in a dose-dependent manner. Conclusion: Serotonin can inhibit the activation of γ-aminobutyric acid.
