目的:探讨辅助生殖技术中巨型卵母细胞的染色体组成以及对于胚胎生长发育的影响。方法:收集40例患者的44枚巨卵细胞,分析了巨卵的成熟度、受精和胚胎发育情况,并采用荧光原位杂交(fluorescence in situ hybridization,FISH)技术分别检测了巨卵及其胚胎的X、13和14号染色体组成。结果:巨卵的发生率约为0.29%,巨卵细胞的平均直径显著大于普通卵母细胞(P<0.05)。获得巨卵的40例患者的平均获卵数明显高于未产生巨卵的患者(P<0.05)。10个巨卵细胞的细胞遗传学检测显示其均为二倍体,对5个2PN巨卵胚胎的13个卵裂球行FISH检测的结果显示,5个胚胎为三倍体或者嵌合体。结论:巨卵受精可能是形成母源性三倍体的原因之一。鉴于其染色体数目异常,对于巨卵发育而来的胚胎可能并不适合用于移植。
Objective: To investigate the chromosomal composition of giant oocytes in assisted reproductive technology and its effect on the growth and development of embryos. Methods: Forty-four giant oocytes were collected from 40 patients. The maturity, fertilization and embryo development of the giant eggs were analyzed. The fluorescence in situ hybridization (FISH) X, 13 and 14 chromosomes. Results: The incidence of giant eggs was about 0.29%, and the average size of giant oocytes was significantly larger than that of normal oocytes (P <0.05). The average number of oocytes retrieved from 40 patients with giant ova was significantly higher than those without ovo (P <0.05). The cytogenetic analysis of 10 giant egg cells showed that they were all diploid. The FISH results of 13 blastomeres from 5 2PN giant egg embryos showed that the three embryos were triploid or chimeric. Conclusion: Giant oocyte fertilization may be one of the reasons for the formation of maternal triploid. Given the anomalous number of chromosomes, embryos derived from giant egg development may not be suitable for transplantation.
