目的探讨水通道蛋白-4(Aquaporin 4,AQP-4)在胰性脑病(Pancreatic encephalopathy,PE)大鼠脑组织损伤过程中的作用。方法 48只SD大鼠,随机分为假手术(Sham-operation,SO)组和胰性脑病(PE)组,每组24只。逆行胰胆管注射5%牛黄胆酸钠(sodium taurocholate,STC)诱导大鼠PE模型,SO组大鼠以生理盐水替代5%STC。比较2组大鼠不同时段脑组织病理学评分、脑内肿瘤坏死因子-α(TNF-α)和AQP-4蛋白及其mRNA表达量,HE染色观察脑组织病理变化、Elisa法检测脑组织中TNF-α和AQP-4含量、实时荧光定量逆转录聚合酶反应(RT-QPCR)检测脑组织中AQP-4-mRNA表达情况。结果与SO组相比,PE大鼠各时间点脑组织病理评分、脑内TNF-α表达量、AQP-4蛋白和AQP-4-mRNA表达量均明显升高(P<0.05),统计发现脑组织病理评分与TNF-α和AQP-4表达量均呈正相关(r=0.761和r=0.659,P均<0.05)。结论 AQP-4在PE大鼠脑组织中表达量增加,推测它在重症急性胰腺炎大鼠并发PE过程中有重要作用。
Objective To investigate the role of aquaporin-4 (AQP-4) in the brain injury induced by pancreatic encephalopathy (PE) in rats. Methods Forty eight Sprague-Dawley rats were randomly divided into sham-operation (SO) group and encephalopathies (PE) group, 24 rats in each group. The rat PE model was induced by injecting 5% sodium taurocholate (STC) into the pancreaticobiliary duct, and the rats in the SO group were given saline instead of 5% STC. The brain histopathological score, the expression of tumor necrosis factor-α (TNF-α) and AQP-4 protein and their mRNA in brain tissue were compared between the two groups at different time points. The pathological changes of brain tissue were observed by HE staining. TNF-α and AQP-4 were detected by real-time fluorescent quantitative reverse transcription polymerase chain reaction (RT-QPCR). Results Compared with SO group, the levels of TNF-α, AQP-4 protein and AQP-4 mRNA in PE rats were significantly increased at different time points (P <0.05) The pathological score of brain tissue was positively correlated with the expression of TNF-α and AQP-4 (r = 0.761 and r = 0.659, P <0.05). Conclusion The expression of AQP-4 in PE rat brain tissue increases, suggesting that it plays an important role in the process of PE complicated by acute pancreatitis in rats.
