目的观察含羞草水提物(WEMP)对人子宫癌细胞(Hela)生长的抑制和凋亡诱导效果及其对mcl-1和bim表达的影响。方法采用MTT比色法观察不同浓度的WEMP对Hela的生长抑制情况;流式细胞仪检测细胞凋亡率;Western blot方法检测WEMP对人胃癌细胞Hela中mcl-1、bim蛋白表达的影响。结果 WEMP作用Hela细胞24 h后,细胞存活率显著降低(P<0.05),且与时间、剂量呈依赖关系。流式细胞仪PI单染亚二倍体峰分析,250,500,1000,2000,4000mg/L WEMP作用Hela细胞24 h,Sub-G1期细胞比率分别为(11.60±0.85)%,(15.95±0.64)%,(18.20±1.41)%、(17.5±0.14)%和(18.10±0.57)%,与对照组(7.4±0.84)%比较差异均有统计学意义(P﹤0.01)。WEMP可降低Hela细胞mcl-1基因表达和增加bim基因表达。结论 WEMP对Hela生长有明显的抑制作用并可诱导Hela的凋亡,mcl-1基因表达降低和bim基因表达增加可能是其凋亡机制之一。
Objective To observe the inhibitory effect of water extract of Mimosa (WEMP) on the growth of human uterine cancer cells (Hela) and the effect of apoptosis induction on the expression of mcl-1 and bim. Methods MTT assay was used to observe the inhibitory effect of different concentrations of WEMP on the growth of Hela cells. Flow cytometry was used to detect the apoptosis rate. Western blot was used to detect the effect of WEMP on the expression of mcl-1 and bim protein in human gastric cancer cell line Hela. Results After treated with WEMP for 24 h, the cell viability was significantly decreased (P <0.05), and dependent on time and dose. Flow cytometry PI single dye sub-diploid peak analysis, 250,500,1000,2000,4000mg / L WEMP Hela cells at 24 h, Sub-G1 phase cells were (11.60 ± 0.85)%, (15.95 ± 0.64) %, (18.20 ± 1.41)%, (17.5 ± 0.14)% and (18.10 ± 0.57)%, respectively, compared with those in control group (7.4 ± 0.84)% (P <0.01). WEMP can reduce the expression of mcl-1 gene and increase the expression of bim gene in Hela cells. Conclusion WEMP can significantly inhibit the growth of Hela cells and induce Hela apoptosis. The decrease of mcl-1 gene expression and the increase of bim gene expression may be one of the mechanisms of apoptosis.
