目的 研究促肾上腺皮质激素释放激素 (Corticotropin releasinghormone ,CRH)对下丘脑神经元胞浆内cAMP浓度和Ca2 + 的调节作用。方法 取孕 17d胎鼠下丘脑分散培养下丘脑神经元 ,采用PTI(Photontechnologyinternational,PTI)测量和分析外源性CRH对培养下丘脑神经元胞浆内游离钙浓度 ( [Ca2 + ]i)变化的影响 ,放射免疫方法测定神经元内cAMP含量。结果 正常对照组的下丘脑神经元 [Ca2 + ]i和cAMP含量较低 ,外源性CRH刺激后 ,[Ca2 + ]i立即升高 ,神经元胞浆内cAMP生成明显增加 ;CP 15 45 2 6可明显抑制CRH ( 10 - 6 mol L)刺激的下丘脑神经元 [Ca2 + ]i和cAMP的生成增加。结论 CRH可通过与其 1受体结合直接作用于下丘脑神经元 ,使神经元 [Ca2 + ]i和cAMP含量明显增加 ,在调节下丘脑神经元激活过程中下丘脑合成和分泌的CRH可能起了重要作用。
Objective To investigate the effects of corticotropin releasing hormone (CRH) on cAMP concentration and Ca2 + in the cytoplasm of hypothalamic neurons. Methods Hypothalamus neurons were disseminated in the hypothalamus of pregnant rats for 17 days. PTI (Photontechnology international, PTI) was used to measure and analyze the change of intracellular free calcium concentration ([Ca2 +] i) in cultured hypothalamus neurons with exogenous CRH Influence, radioimmunoassay method to determine cAMP content in neurons. Results The content of [Ca2 +] i and cAMP in the hypothalamus neurons was lower in the normal control group. [Ca2 +] i increased immediately after exogenous CRH stimulation and the cAMP production in the neurons increased significantly. CP 15 45 2 6 significantly inhibited the increase of [Ca2 +] i and cAMP production in hypothalamic neurons stimulated by CRH (10 - 6 mol L). CONCLUSIONS: CRH directly interacts with hypothalamic neurons by binding to its receptor 1 and significantly increases the contents of [Ca2 +] i and cAMP in the hypothalamus. CRH may play an important role in regulating hypothalamic synthesis and secretion during hypothalamic neuronal activation Important role.
