为了验证亚比棉×陆地棉杂种F1的真实性,从细胞学和RAPD技术方面对亚比棉×陆地棉杂种F1及其亲本进行了研究。细胞学研究表明,三种杂种(亚洲棉×比克氏棉)×陆地棉的核型公式为2n=4x=52=2M+44m(2SAT)+6sm(2SAT),相对长度变异范围为6.12%~2.56%,最长染色体与最短染色体之比为2.39∶1,臂比值在1.00~1.82之间,平均臂比为1.35,核型类别为1B。RAPD分析表明,亚比棉×陆地棉杂种F1共扩增出61条DNA带,其中具亚洲棉特异带的占总带数的3.3%,来自比克氏棉的带占9.8%,具陆地棉特异带的占13.1%,来自亚比棉的特异带占9.8%,杂种的特异带占14.8%。细胞学研究和RAPD分析的结果吻合,从而断定亚比棉×陆地棉杂种F1为真正的杂交种。
In order to verify the authenticity of F1 hybrids between cotton and cotton cultivars, the hybrids F1 and their parents were analyzed from the aspects of cytology and RAPD. Cytological studies showed that the karyotypes of three hybrids × cotton × cotton × Upland cotton were 2n = 4x = 52 = 2M + 44m (2SAT) + 6sm (2SAT), and the relative length ranged from 6.12% ~ 2.56%, the ratio of the longest chromosome to the shortest chromosome was 2.39:1, the arm ratio was between 1.00 and 1.82, the average arm ratio was 1.35, and the karyotype was 1B. RAPD analysis showed that 61 DNA bands were amplified by hybrid F1 of cotton × cotton accessions, including 3.3% of the total number of tapes in Asia, 9.8% of the total number of tapes, 13.1% of the specific bands, 9.8% of the specific bands from the cotton and 14.8% of the hybrids. Cytological studies and RAPD analysis of the results of the agreement, thus concluded that cotton than cotton × Upland cotton hybrid F1 as a true hybrid.
