目的探讨自噬基因Beclin 1在甲状腺乳头状癌(PTC)组织中的表达及其临床意义。方法应用免疫组织化学SP法,检测90例甲状腺乳头状癌组织和60例癌旁正常甲状腺组织中Be-clin 1的表达,分析Beclin 1的表达与PTC生物学行为的关系。结果 Beclin 1在60例癌旁正常甲状腺组织中有45例表达阳性,阳性表达率为75.0%,而在90例PTC组织中表达阳性18例,阳性表达率为20.0%,明显低于癌旁正常甲状腺组织中的表达(2=8.24,P<0.01),Beclin 1在90例PTC组织中的表达与是否有颈部淋巴结转移和浸润深度密切相关(P<0.01)。结论 Beclin 1表达缺失可能是PTC形成过程中的重要机制之一,可能成为探讨肿瘤发生和抑制肿瘤浸润转移研究的新靶点。
Objective To investigate the expression of autophagy gene Beclin 1 in papillary thyroid carcinoma (PTC) and its clinical significance. Methods Immunohistochemical SP method was used to detect the expression of Be-clin 1 in 90 cases of thyroid papillary carcinoma and 60 cases of normal thyroid tissues. The relationship between the expression of Beclin 1 and the biological behavior of PTC was analyzed. Results The expression of Beclin 1 in 45 cases of normal thyroid tissue adjacent to cancer was 45.0%, while in 90 cases of PTC tissues, the positive rate was 20.0%, which was significantly lower than that in adjacent normal tissues (2 = 8.24, P <0.01). The expression of Beclin 1 in 90 cases of PTC was closely related to the lymph node metastasis and depth of invasion (P <0.01). Conclusion The loss of Beclin 1 expression may be one of the important mechanisms in the process of PTC formation, which may be a new target for the study of tumorigenesis and suppression of tumor invasion and metastasis.
