目的:研究抗酸和免疫组织化学双重染色法在同一张切片上清晰显示结核分支杆菌与巨噬细胞的分布状态。方法:组织切片常规脱蜡、入水,其染色分别按抗酸-免疫组化-苏木素;免疫组化-抗酸-苏木素和免疫组化-苏木素-抗酸顺序进行。结果:组织切片按免疫组化-抗酸-苏木素顺序染色,背景清晰,反差明显,较另两种染色顺序显色效果好。结论:组织切片按首先免疫组织化学染色,在抗酸染色,最后苏木素染色的步骤染色效果最好。能在同一切片中清晰显示结核分枝杆菌与巨噬细胞的分布情况。
OBJECTIVE: To study the distribution of Mycobacterium tuberculosis and macrophages in the same slice by double-stained antacid and immunohistochemical staining. Methods: Tissue sections were deparaffinized and infused into water. Staining was performed according to antacid - immunohistochemistry - hematoxylin; immunohistochemistry - antacid - hematoxylin and immunohistochemistry - hematoxylin - antacid sequencing. Results: Tissue sections were stained immunohistochemically-antacid-hematoxylin sequence with clear background and obvious contrast, which was more effective than the other two staining sequences. Conclusion: Tissue sections were stained immunohistochemically first and stained best in the steps of acid-fast staining and final hematoxylin-stained staining. Can be clearly displayed in the same section of the distribution of Mycobacterium tuberculosis and macrophages.
