目的:研究酶法转化制备虎杖中白藜芦醇的最佳工艺。方法:以酶解液中白藜芦醇含量为指标,通过正交实验筛选出合适的配比酶制剂,确定了酶解条件。结果:最佳配比酶用量为SPE-002为0.20%、SPE-005为0.17%、SPE-007为0.08%;最佳酶解条件为温度40℃、pH5.0、时间120min、底物浓度15%。结论:通过酶解使白藜芦醇苷转化为白藜芦醇,产物含量由1.44mg/g提高到14.16mg/g。
Objective: To study the best enzymatic conversion of resveratrol in Polygonum cuspidatum. Methods: The content of resveratrol in enzymatic hydrolyzate was used as an index, and the suitable ratio enzyme preparation was screened by orthogonal experiment, and the conditions of enzymolysis were determined. Results: The optimum amount of enzyme was 0.20% for SPE-002, 0.17% for SPE-005 and 0.08% for SPE-007. The optimal conditions of enzymatic hydrolysis were temperature40 ℃, pH5.0, time120min, substrate concentration 15%. Conclusion: Polydatin was converted to resveratrol by enzymatic hydrolysis, and the content of the product increased from 1.44 mg / g to 14.16 mg / g.
