[目的]探讨人脑胶质瘤组织中EphA2和MMP2的表达水平及相关性。[方法]采用免疫组织化学SP法检测69例胶质瘤组织、10例正常脑组织中EphA2和MMP2蛋白表达情况,分析两者表达水平与胶质瘤临床病理分级之间的关系及两者的相关性。[结果]正常脑组织、低级别胶质瘤组织(Ⅰ~Ⅱ级)和高级别胶质瘤组织(Ⅲ~Ⅳ级)中EphA2蛋白表达的阳性率分别为10.0%(1/10)、37.0%(10/27)、90.5%(38/42),差异有统计学意义(χ2=33.0826,P=0.000);正常脑组织、低级别胶质瘤组织和高级别胶质瘤组织中MMP2蛋白表达的阳性率分别为0、14.8%(4/27)、69.0%(29/42),差异有统计学意义(χ2=28.0869,P=0.000)。在胶质瘤组织中,EphA2与MMP2蛋白表达呈正相关(r=0.660,P=0.000)。[结论]EphA2与MMP2同时高表达于胶质瘤组织中,EphA2可能通过影响MMP2而促进肿瘤的侵袭与转移。
[Objective] To investigate the expression and correlation of EphA2 and MMP2 in human glioma tissue. [Methods] The expression of EphA2 and MMP2 in 69 glioma tissues and 10 normal brain tissues were detected by immunohistochemical SP method. The relationship between the expression of EphA2 and MMP2 and the clinicopathological grade of gliomas was analyzed. Correlation. [Results] The positive rates of EphA2 protein expression in normal brain tissue, low grade glioma tissue (Ⅰ ~ Ⅱ grade) and high grade glioma tissues (Ⅲ ~ Ⅳ grade) were 10.0% (1/10), 37.0 % (10/27), 90.5% (38/42) respectively. The difference was statistically significant (χ2 = 33.0826, P = 0.000). In normal brain tissue, low grade glioma tissue and high grade glioma tissue, MMP2 protein The positive rates were 0,14.8% (4/27) and 69.0% (29/42), respectively. The difference was statistically significant (χ2 = 28.0869, P = 0.000). In glioma tissue, EphA2 and MMP2 protein expression was positively correlated (r = 0.660, P = 0.000). [Conclusion] Both EphA2 and MMP2 are highly expressed in glioma tissues. EphA2 may promote the invasion and metastasis of tumor by affecting MMP2.
