目的 探讨乳腺癌组织层粘连蛋白 (LN)及其受体 (LN R)和nm2 3蛋白的表达与间质微血管密度及肿瘤转移的关系。方法 应用免疫组织化学LSAB方法检测 73例乳腺癌LN ,LN R和nm2 3蛋白的表达 ,在第Ⅷ因子相关抗原 (FⅧRAg)染色切片上检测其微血管密度 (MVD)。结果 病理分级越高 ,乳腺癌LN表达程度越强 ,即病理Ⅰ级而LN分布Ⅲ级者为 8.3% ,病理和LN分布均Ⅲ级者为 5 8.8% ,两者间差异有极显著意义。LN R表达程度则与转移有关 ,转移组表达程度 (78.6 % )高于非转移组 (2 6 .7% )。LN R的表达程度与乳腺癌间质MVD呈正相关。淋巴结转移率 ,nm2 3表达阳性组与阴性组比较差异有显著性意义。在nm2 3表达抑制时 ,LN R ,MVD增加 ,淋巴结转移明显增加。结论 LN R表达增强 ,MVD增加 ,nm2 3表达抑制 ,可能作为预测肿瘤转移及判断预后的可靠指标
Objective To investigate the relationship between the expression of laminin (LN), its receptor (LN R) and nm23 protein and the interstitial microvessel density and tumor metastasis in breast cancer. Methods The expression of LN, LN R and nm23 protein in 73 cases of breast cancer was detected by immunohistochemistry LSAB method. Microvessel density (MVD) was detected on VIII factor-associated antigen (FVIIIRAg) stained sections. Results The higher the pathological grade was, the stronger the degree of LN expression in breast cancer was, that is, pathological grade I and LN distribution grade III were 8.3%, pathological and LN distribution grade III were 5 8.8%, the difference between the two was extremely significant. The degree of LN R expression was related to metastasis. The degree of expression in the metastatic group (78.6%) was higher than that in the non-metastatic group (26.7%). The expression of LN R was positively correlated with the MVD of breast cancer. The lymph node metastasis rate was significantly higher in the nm23 positive group than in the negative group. In the inhibition of nm23 expression, LN R, MVD increased, lymph node metastasis increased significantly. Conclusion The expression of LN R is increased, MVD is increased, and the expression of nm23 is inhibited. It may be a reliable index for predicting tumor metastasis and judging prognosis.
