目的 探索 Sj CTPI和 Sj C2 3DNA疫苗联合应用保护性免疫的协同作用。方法 大量制备 pc DNA3.1- Sj CTPI和 pc DNA3.1- Sj C2 3的质粒 DNA。 4 8只 BAL B/ c小鼠分为 4个组 (A、B、C、D组 )每组 12只。 A组每鼠经双侧股四头肌注射 10 0 μg pc DNA3.1的质粒 DNA,为对照组 ;B组为Sj C2 3组 ,每鼠肌注 10 0μg pc DNA3.1- Sj CTPI的质粒 DNA;C组为 Sj CTPI组 ,每鼠肌注 10 0μgpc DNA3.1- Sj CTPI的质粒 DNA;D组为联合免疫组 ,每组肌注 10 0μg pc DNA3.1- Sj C2 3和 10 0μgpc DNA3.1- Sj CTPI质粒 DNA的混和物。分别于 0、14、2 8d免疫 3次。末次免疫后 30 d,每鼠经腹部皮肤攻击感染 (45± 2 )条日本血吸虫尾蚴。攻击后 4 5 d,剖杀所有小鼠 ,灌冲 ,收集成虫并计数。每鼠肝脏称重后剪碎 ,置 5 % KOH中消化 ,并计数虫卵数。比较各组间的减虫率和减卵率。结果 与对照组相比 ,Sj C2 3组、Sj CTPI及联合组的减虫率分别为 2 8.1%、2 9.1%和 4 1.5 % ,均非常显著地高于对照组 (P分别为 <0 .0 1,<0 .0 1,<0 .0 0 1) ,而联合组则又显著地高于单独 Sj C2 3组及单独 Sj CTPI组 (P均 <0 .0 5 )。三组的减卵率分别为 37.9%、4 4 .2 %和 6 1.4 % ,均非常显著地高于对照组 (P分别为 <0 .0 1,<0 .0 1,<0 .0 0 1) ,但?
Objective To explore the synergistic effect of combined immunization with Sj CTPI and Sj C2 3DNA vaccines. Methods Plasmid DNAs of pcDNA3.1-Sj CTPI and pc DNA3.1-Sj C2 3 were prepared in large quantities. Forty eight BALB / c mice were divided into four groups (Groups A, B, C and D) with 12 mice in each group. In group A, plasmid DNA of 100 μg pcDNA3.1 was injected into the quadriceps of both sides of the quadriceps muscle in each group, as a control group; in group B, Sj C2 3 group was injected with 10 μg pcDNA3.1-Sj CTPI plasmid DNA; C group Sj CTPI group, intramuscular injection of 10μg pcDNA3.1-Sj CTPI plasmid DNA; D group for the combined immunization group, each group of intramuscular injection of 10μg pcDNA3.1-Sj C2 3 and 10μg pcDNA3 Mixtures of 1-Sj CTPI plasmid DNA. Were immunized at 0,14,2 8d three times respectively. Thirty days after the last immunization, each mouse was challenged by abdominal skin challenge with (45 ± 2) cercariae of Schistosoma japonicum. On day 4 after challenge, all mice were sacrificed, infused, and adults collected and counted. The liver of each rat was weighed, cut into pieces, digested in 5% KOH, and the number of eggs counted. The worm reduction rate and the oviposition rate were compared between groups. Results Compared with the control group, the worm reduction rates of Sj C2 3 group, Sj CTPI group and the combined group were 21.1%, 21.9% and 41.5%, respectively, which were significantly higher than those in the control group (P <0. 0 1, <0 1 0 1 0 0 0 1), while the combined group was significantly higher than Sj C2 3 alone group and Sj CTPI alone group (P <0.05).
