目的 研究伤寒杆菌耐药质粒 pRST98与细菌外膜泡 (OMV)形成的关系。方法 将 ( 1)携带 pRST98的野生型耐药伤寒杆菌———ST1及ST2 ;( 2 )不含 pRST98的野生型敏感伤寒杆菌———ST3及ST4 ;( 3 )不含质粒的大肠杆菌———E .coliK12 W14 85;( 4)用十二烷基硫酸钠消除 pRST98的突变体菌株———ST1(R-)及ST2 (R-) ;和 ( 5 )经接合转移将pRST98导入敏感型伤寒杆菌及大肠杆菌的接合子———pRST98/ST3、pRST98/ST4 及pRST98/E .coliK12 W14 85用于实验 ,用透射电镜观测细菌在有无 pRST98时 ,OMV形成的差异。结果 携带 pRST98的耐药伤寒杆菌消除 pRST98前后 ,不含 pRST98的敏感伤寒杆菌和大肠杆菌在导入该质粒前后 ,细菌形成OMV的数量和形态均无明显变化。结论 伤寒杆菌耐药质粒pRST98在体外与细菌外膜泡的形成未发现有明显关系
Objective To study the relationship between typhimurium resistant plasmid pRST98 and bacterial outer membrane vesicle (OMV). Methods (1) wild-type resistant Salmonella typhi - ST1 and ST2 carrying pRST98; (2) wild-type Salmonella typhimurium - ST3 and ST4 without pRST98; (3) plasmid- - E. coli K12 W14 85; (4) elimination of mutant strains of pRST98 - ST1 (R-) and ST2 (R-) with sodium dodecyl sulfate; Salmonella typhi and E. coli conjugates --- pRST98 / ST3, pRST98 / ST4 and pRST98 / E. ColiK12 W14 85 were used in the experiment to observe the differences in the formation of OMV in bacteria with or without pRST98 by transmission electron microscopy. Results The number and morphology of OMV formed by bacterium did not change before and after the introduction of pRST98 and pRST98-resistant typhoid bacillus and E. coli without pRST98. Conclusion Salmonella typhi resistance plasmid pRST98 was not found to have a significant relationship with the formation of bacterial outer membrane vesicles in vitro
