目的:检测卵巢癌构域蛋白酶1(ovarian tumor domain-containing protease 1,OTUD1)在非小细胞肺癌(non-small cell lung cancer,NSCLC)组织中的表达水平,探讨OTUD1表达与NSCLC临床病理特征及预后的关系。方法:选取2008年3月至2009年2月在山东大学齐鲁医院胸外科行手术治疗的82例非小细胞肺癌患者组织标本及40例正常肺组织标本,免疫组化法检测OTUD1的表达水平;另取2015年3月至2015年4月在山东大学齐鲁医院胸外科手术治疗的原发性NSCLC 25例,应用Real-time PCR检测OTUD1在NSCLC组织及邻近正常肺组织内的表达。结果:与正常肺组织相比,NSCLC组织中OTUD1 mRNA的表达显著增高(0.620±0.055 vs 0.387±0.037,P<0.01);OTUD1的表达与患者的年龄、性别、吸烟史、病理类型均无显著相关性(均P>0.05);与淋巴结转移(P<0.01)、分化程度(P<0.01)及TNM分期(P<0.05)显著相关。单因素及多因素分析结果显示,OTUD1高表达与患者不良预后有关。结论:OTUD1在NSCLC组织中高表达,与NSCL的恶性进展及不良预后密切相关,可作为判断NSCLC生物学特征及预后的参考指标。
OBJECTIVE: To detect the expression of OTUD1 in non-small cell lung cancer (NSCLC) and to investigate the clinicopathological characteristics of OTUD1 and NSCLC. Prognosis of the relationship. Methods: From March 2008 to February 2009 in Shandong Qilu Hospital thoracic surgery in 82 cases of non-small cell lung cancer patients and 40 cases of normal lung tissue specimens, immunohistochemical detection of OTUD1 expression levels; In addition, 25 patients with primary NSCLC treated with thoracic surgery at Qilu Hospital of Shandong University from March 2015 to April 2015 were selected. The expression of OTUD1 in NSCLC tissues and adjacent normal lung tissues was detected by Real-time PCR. Results: The expression of OTUD1 mRNA in NSCLC tissues was significantly higher than that in normal lung tissues (0.620 ± 0.055 vs 0.387 ± 0.037, P <0.01). The expression of OTUD1 was not associated with age, sex, smoking history and pathological type (P <0.01), differentiation degree (P <0.01) and TNM stage (P <0.05). Univariate and multivariate analysis showed that high OTUD1 expression correlated with poor prognosis. Conclusion: The high expression of OTUD1 in NSCLC tissues is closely related to the malignant progression and poor prognosis of NSCL, which may be used as a reference index to judge the biological characteristics and prognosis of NSCLC.
