用肺腺癌细胞系A549免疫BALB/C小鼠,常规细胞融合,获得一株ⅠgM型单抗3C9。westernblot研究发现该抗体识别56.4KD蛋白带。免疫组化研究发现,3C9相关抗原(3C9Ag)在肺癌组织中表达阳性率高达92.5%。3C9Ag血清学研究发现,其诊断肺癌的敏感性为64.7%,特异性为93.0%,准确性为82.5%。其血清水平与组织学分布相关,在高、中分化的肺癌病人阳性率高于低分化者(P<0.05)。表明检测血清3C9Ag具有重要意义。
The lung adenocarcinoma cell line A549 was used to immunize BALB/C mice and routine cell fusion was performed to obtain a IgM type monoclonal antibody 3C9. Westernblot study found that this antibody recognizes 56.4KD protein band. Immunohistochemical study found that the positive rate of 3C9-associated antigen (3C9Ag) in lung cancer tissues was as high as 92.5%. 3C9Ag serological studies found that its sensitivity for the diagnosis of lung cancer was 64.7%, specificity was 93.0%, and accuracy was 82.5%. The serum levels were correlated with histological distribution. The positive rate of patients with high and moderately differentiated lung cancer was higher than those with poor differentiation (P<0.05). It shows that the detection of serum 3C9Ag is of great significance.
