目的观察成熟耳石破坏再生过程中,Otoconin 90(OC90)蛋白在内耳的分布表达情况,探讨其在成熟耳石破坏后修复再生过程中的意义。方法采用机械过载作用的方法造成豚鼠耳石破坏模型,免疫组织化学检测耳石破坏后不同时期(0.5~1 h,3 d,7 d)内耳切片各主要部位OC90分布表达情况,并测定椭圆囊囊斑的光密度(OD)值,对其表达强度进行半定量分析。结果 OC90蛋白的分布在耳石破坏后没有显著变化,仍呈现“多点”表达的特点,其在椭圆囊和球囊囊斑感觉上皮的表达于破坏后呈现增强趋势,其中耳石破坏3 d时的OD值增加显著(P﹤0.05)。结论 OC90蛋白在成熟耳石破坏后修复再生过程中起重要作用。
Objective To observe the distribution of Otoconin 90 (OC90) protein in the inner ear during the regeneration and regeneration of mature otolith, and to explore the significance of Otoconin 90 (OC90) protein in the process of regeneration and regeneration after the destruction of mature otolith. Methods The model of otolith disruption was induced by mechanical overload in rats. Immunohistochemistry was used to detect the expression of OC90 in various sections of the inner ear at various stages (0.5-1 h, 3 d, 7 d) after otolith disruption. The optical density (OD) value of the plaque was used to semi-quantitatively analyze its expression intensity. Results The distribution of OC90 protein did not change significantly after otolith disruption, and still showed the expression of “multi-point” expression. The expression of OC90 protein showed an increasing trend after the destruction of the oval and balloon punctate sensory epithelium, in which otolith destruction The OD at 3 d increased significantly (P <0.05). Conclusion OC90 protein plays an important role in the repair and regeneration of mature otolith.
