目的调查多药耐药鲍氏不动杆菌(MDR-ABA)转座子、整合子等可移动遗传元件的存在情况。方法收集宁波市李惠利医院2009年6-9月住院患者中分离的MDR-ABA共20株,采用聚合酶联反应(PCR)及序列分析的方法分析10种可移动遗传元件:tnpU、tnp513、IS26、ISEcp1、ISaba1、ISaba4、ISaba9、intⅠ1、intⅠ2、intⅠ3。结果 20株MDR-ABA共检出5种基因:tnpU、tnp513、IS26、ISaba1、intⅠ1,其余5种基因未检出;各种基因阳性率以IS26和ISaba1最高16株(80%),intⅠ1次之13株(65%),tnp513为12株(60%),tnpU9株(45%);9株MDR-ABA同时检出5种基因,1株MDR-ABA同时检出4种基因,2株MDR-ABA同时检出3种基因,5株MDR-ABA同时检出2种基因,1株MDR-ABA仅检出1种基因,另有2株MDR-ABA未检出上述10种基因。结论同时检测MDR-ABA10种可移动遗传元件是国内首次报道,可移动遗传元件介导各种耐药基因使受体菌表现为多药耐药。
Objective To investigate the existence of mobile genetic elements such as multi-drug resistant Acinetobacter baumannii (MDR-ABA) transposons and integrons. Methods A total of 20 MDR-ABA strains isolated from hospitalized patients in Li Hui Li Hospital of Ningbo City from June to September in 2009 were collected and 10 mobile genetic elements including tnpU, tnp513 and tnp513 were analyzed by polymerase chain reaction (PCR) and sequence analysis. IS26, ISEcp1, ISaba1, ISaba4, ISaba9, intI1, intI2, intI3. Results Five genes were detected in 20 MDR-ABA strains: tnpU, tnp513, IS26, ISaba1 and intⅠ1, and the other 5 genes were not detected. The positive rate of each gene was 16 (80%) in IS26 and ISaba1, 13 strains (65%), 12 strains (60%) were tnp513 and 45 strains were tnpU9 (45%). Nine genes were detected in 9 strains of MDR-ABA, 4 genes were detected in one strain of MDR- Three genes were detected at the same time in MDR-ABA. Two genes were detected simultaneously in five MDR-ABA genes. Only one gene was detected in MDR-ABA and the other two genes were not detected in two MDR-ABA genes. Conclusions Simultaneous detection of 10 MDR-ABA mobile genetic elements is the first reported in China. The mobile genetic elements mediate various resistance genes and render the recipient bacteria multi-drug resistant.
