目的探讨胰岛新生相关蛋白肽(Islet neogenesis associated protein-pp,INGAP-pp)对体外培养的人胰岛细胞分泌胰岛素水平的影响。方法取成人胰头癌患者手术中切除的胰腺组织正常胰尾部分,分离纯化胰岛进行体外培养,检测胰岛的生物学活性,并比较添加与不添加INGAP-pp培养的胰岛分泌胰岛素能力的变化。结果分离纯化的胰岛具有良好的生物学活性。在体外培养21 d后,未添加INGAP-pp的胰岛分泌胰岛素的能力逐渐丢失,胰岛形态发生显著变化,细胞团崩解后细胞逐渐死亡;而添加了INGAP-pp的胰岛形态保持相对良好,且胰岛素分泌水平从培养的第6天起均显著高于未添加组(P<0.05或P<0.01)。结论 INGAP-pp能显著促进体外培养的人胰岛细胞分泌胰岛素。
Objective To investigate the effect of islet neogenesis associated protein-pp (INGAP-pp) on the insulin secretion in cultured human pancreatic islet cells. Methods The normal pancreatic tail part of pancreatic tissue removed from the patients with pancreatic head carcinoma was isolated and purified. The islets were isolated and purified for in vitro culture, and the biological activity of islets was measured. The insulin secretion ability of islets cultured with and without INGAP-pp was compared. Results Isolation and purification of islets have good biological activity. After cultured for 21 days in vitro, the ability of islets to release insulin without INGAP-pp was gradually lost, the morphology of islets changed significantly, and the cells gradually died after disintegration of cells; while the islet morphology with INGAP-pp remained relatively good, and The level of insulin secretion from the sixth day of culture was significantly higher than that of the non-added group (P <0.05 or P <0.01). Conclusion INGAP-pp can significantly promote the insulin secretion in cultured human islet cells.
