目的:探讨氧化型低密度脂蛋白(ox-LDL)刺激人单核/巨噬细胞对Notch1表达的影响。方法:将人单核细胞株(THP1)经氟波酯(PMA)刺激转化为巨噬细胞后给予不同浓度ox-LDL刺激,采用RT-PCR及Westernblot法检测巨噬细胞中Notch1受体mRNA及蛋白水平的表达,采用RT-PCR及ELISA法测定血管细胞黏附分子1(VCAM-1)和单核细胞趋化因子1(MCP-1)的表达。结果:与对照组比较,ox-LDL能显著上调巨噬细胞中Notch1受体mRNA及蛋白水平的表达(P<0.05),增加促炎细胞因子VCAM-1和MCP-1的分泌,并在一定范围内与ox-LDL呈浓度相关性。结论:ox-LDL能够激活巨噬细胞Notch1信号,增加促炎细胞因子分泌,促进巨噬细胞活化。
AIM: To investigate the effect of ox-LDL on Notch1 expression in human monocytes / macrophages. Methods: Human monocyte strain (THP1) was stimulated with fluvastatin (PMA) into macrophages and stimulated with different concentrations of ox-LDL. The expression of Notch1 receptor mRNA and protein in macrophages were detected by RT-PCR and Western blotting. The protein expressions of VCAM-1 and MCP-1 were determined by RT-PCR and ELISA. Results: Compared with the control group, ox-LDL significantly up-regulated the mRNA and protein expression of Notch1 in macrophages (P <0.05) and increased the secretion of pro-inflammatory cytokines VCAM-1 and MCP-1 Concentration was correlated with ox-LDL. Conclusion: Ox-LDL can activate Notch1 signal of macrophages, increase the secretion of proinflammatory cytokines and promote the activation of macrophages.
