目的探讨d-δ-三烯生育酚抗鼻咽癌的生物学活性。方法将低分化5-8F鼻咽癌细胞随机分为观察组和对照组,观察组分别予40、50、60μmol/L d-δ-三烯生育酚干预48 h,对照组不予干预。采用MTT实验、流式细胞仪检测细胞增殖抑制率、凋亡率及细胞周期;荧光染色和显微镜观察细胞形态学变化。结果观察组干预48 h后,随浓度增加,镜下观察活细胞减少,漂浮的死亡细胞增多,细胞核强荧光染色细胞比例增多,细胞出现明显的抑制和凋亡现象。观察组不同浓度细胞抑制率及凋亡率明显升高,且明显高于对照组(P均<0.01);对照组S期的细胞百分率明显低于观察组,P均<0.01。结论 d-δ-三烯生育酚能抑制5-8F细胞增殖,诱导其凋亡,具有潜在的抗鼻咽癌生物学活性。
Objective To investigate the biological activity of d-δ-tocotrienols against nasopharyngeal carcinoma. Methods The poorly differentiated 5-8F NPC cells were randomly divided into observation group and control group. The observation group were treated with 40, 50, 60μmol / L d-δ-tocotrienol for 48 hours without any intervention in the control group. MTT assay was used to detect cell proliferation inhibition rate, apoptosis rate and cell cycle by flow cytometry. Fluorescence staining and microscopy were used to observe the cell morphological changes. Results After 48 hours of intervention, the number of viable cells decreased, the number of floating cells increased, the proportion of strongly stained nuclei increased, and the cells showed obvious inhibition and apoptosis as the concentration increased. The cell inhibitory rate and apoptosis rate in observation group were significantly higher than those in control group (P <0.01). The percentage of cells in control group at S phase was significantly lower than that in observation group (P <0.01). Conclusion d-δ-tocotrienol can inhibit the proliferation of 5-8F cells and induce its apoptosis, which has potential biological activity against nasopharyngeal carcinoma.
