目的研究miRlet7b在氧糖剥夺诱导原代Wistar大鼠增殖星形胶质细胞的表达,探讨缺氧缺血性脑损伤后星形胶质细胞增殖的分子机制。方法原代Wistar大鼠星形胶质细胞随机分为实验组(氧糖剥夺诱导的增殖组)和对照组(正常培养组)。EdU掺入法检测两组细胞的增殖,Western blot检测VEGF蛋白表达,real-time PCR方法检测两组细胞miRlet7b mRNA的表达。结果实验组EdU阳性细胞率,VEGF蛋白表达水平显著高于对照组(P<0.01),而实验组miRlet7b mRNA的表达水平显著低于对照组(P<0.01)。结论 MiRlet7b mRNA表达下调可能是缺氧缺血性脑损伤后星形胶质细胞增殖的分子机制之一,上调VEGF可能促进星形胶质细胞增殖。
Objective To investigate the expression of miRlet7b in primary Wistar rat astrocyte induced by oxytocin and explore the molecular mechanism of astrocyte proliferation after hypoxic-ischemic brain damage. Methods Primary Wistar rats astrocytes were randomly divided into experimental group (oxygen glucose deprivation induced proliferation group) and control group (normal culture group). EdU incorporation method was used to detect the proliferation of the two groups of cells. The protein expression of VEGF was detected by Western blot. The expression of miRlet7b mRNA was detected by real-time PCR. Results The positive rate of EdU cells and the expression of VEGF protein in the experimental group were significantly higher than those in the control group (P <0.01). However, the expression of miRlet7b mRNA in the experimental group was significantly lower than that of the control group (P <0.01). Conclusions The down-regulation of MiRlet7b mRNA may be one of the molecular mechanisms of astrocyte proliferation after hypoxic-ischemic brain damage. Upregulation of VEGF may promote the proliferation of astrocytes.
